福氏志贺菌ipaB基因的克隆表达及免疫原性研究 被引量:1

摘要 目的:构建福氏志贺菌ipaB基因的原核表达载体,诱导表达并纯化。探讨重组蛋白IpaB经口服免疫小鼠后,机体产生的免疫应答。方法:用PCR扩增ipaB目的基因,克隆至pQE-30表达载体中,转化宿主菌DH5α,经IPTG诱导表达蛋白IpaB;Western blot分析其抗原性。纯化后灌胃免疫小鼠,ELISA法检测血清IgG、IgA、小肠黏液sIgA和粪便sIgA。结果:ipaB基因全长1743bp。经SDS-PAGE分析相对分子量(Mr)约为64000。Western blot分析能与福氏志贺菌全菌抗血清反应。免疫小鼠后,血清IgA、IgG,粪sIgA,肠黏液sIgA明显高于对照组(P<0.01)。结论:成功构建了ipaB基因的原核表达载体并能高效表达,纯化后口服免疫可有效诱导黏膜免疫应答,产生高水平的sIgA,发挥局部黏膜免疫作用,为进一步研究细菌性痢疾的疫苗和发病机制奠定基础。
