摘要
目的观察活血解毒方对糖尿病大鼠坐骨神经中糖基化终产物(Glycation end products,AGEs)-糖基化终产物受体(Glycation end product receptor,RAGE)-核因子-κB(nuclear factor-B,NF-κB)途径的影响。方法雄性SD大鼠分为正常组、模型组、甲钴胺组、活血解毒方高、低剂量组。链脲佐菌素(streptozocin,STZ)腹腔注射65mg/kg造模,免疫组化法检测坐骨神经中AGEs、RAGE、NF-κB蛋白的表达。结果与正常组比较,模型组大鼠坐骨神经内AGEs、RAGE、NF-κB水平升高;与模型组比较,活血解毒方组大鼠坐骨神经内AGEs、RAGE、NF-κB表达降低。结论活血解毒方可能是通过抑制AGEs-RAGE-NF-κB途径从而防治糖尿病神经病变。
Objective To investigate the effect of Huoxuejiedu formula on AGEs-RAGE-NF-κB pathway in diabetic rats.Methods Male SD rats were divided into normal group,model group,methylcobalamin group,Huoxuejiedu formula group.The diabetic rats were established by streptozocin injected intra-peritoneally.The 16-week treatment started after confirmation of diabetes,the expression of AGEs,RAGE and NF-κB in sciatic nerve were detected using immunohistochemistry.Results Expression of AGEs,RAGE and NF-κB protein were increased in model group compared with normal group.The expression of AGEs,RAGE and NF-κB were reduced in Huoxuejiedu formula group.Conclusion Huoxuejiedu formula prevents diabetic neuropathy by inhibiting AGEs-RAGE-NF-κB pathway.
