摘要
目的观察蟛蜞菊内酯对脂多糖(lipopolysaccharide,LPS)诱导RAW264.7细胞IL-6、TNF-α及NF-κB的作用。方法酶联免疫吸附试验(ELISA)检测0.2,2和20μmol.L-1低、中、高3浓度蟛蜞菊内酯对终浓度为10μg.ml-1LPS诱导RAW264.7细胞产生白细胞介素-6(IL-6)及肿瘤坏死因子-α(TNF-α)的影响,Western blot方法检测蟛蜞菊内酯对LPS诱导NF-κB细胞核因子-κB(NF-κB)p65蛋白表达的影响。结果 LPS能够明显诱导小鼠RAW264.7细胞产生的IL-6、TNF-α产生,蟛蜞菊内酯低、中、高3个浓度均能抑制LPS诱导产生的的IL-6、TNF-α产生,呈现剂量依赖性。小鼠RAW264.7细胞株产生的NF-κB p65表达可以被LPS诱导增加,与空白组比有显著差异。蟛蜞菊内酯低中高3个浓度均能抑制LPS诱导产生的NF-κB p65表达。结论蟛蜞菊内酯可以抑制LPS所致的RAW264.7细胞炎症反应,其抗炎作用与抑制NF-κB活化,进而减少炎性细胞因子IL-6、TNF-α的生成有关。
Objective To study the effect of Wedelolactone on IL-6、TNF-α and NF-κB p65 expression in lipopolysaccharide(LPS) induced RAW264.7 cells.Methods The contents of IL-6 and TNF-α were detected by ELISA methods.The expression of NF-κB p65 protein in RAW264.7 cells was measured by Western blot,in the presence or absence of 10 μg.ml-1 lipopolysaccharide(LPS) or Wedelolactone(0.2,2,20μmol.L-1).Results Compared with control group,the contents of IL-6 and TNF-α were significantly induced by LPS,Wedelolactone inhibited the expression of IL-6 and TNF-α significantly elicited by LPS in RAW264.7 cells,in dose dependent manners.LPS-induced the expression of NF-κB p65 protein was significantly inhibited by Wedelolactone in RAW264.7 cells.Conclusion Wedelolactone has anti-inflammatory effect,which might be mediated by down-regulating inflammatory factors IL-6,TNF-α through NF-κB signal pathway.
