复合螺旋藻多糖对人乳腺癌MB-231细胞株体外抗肿瘤作用研究 被引量:4

摘要 目的:研究复合螺旋藻多糖对体外培养的人乳腺癌MB-231肿瘤细胞的抑制作用及可能的作用机制。方法:以人乳腺癌MB-231细胞株为研究对象,将螺旋藻多糖(PSP)与银杏叶提取物(GBE)按不同比例复合,并用不同浓度的复合制剂分别对细胞作用后,分别在48h、72h后,显微镜下观察细胞形态学变化,MTT法测其生长抑制率。48h后,PI及AnnexinV-FITC/PI染色,用流式细胞仪测其细胞周期及凋亡率。结果:复合螺旋藻多糖对人乳腺癌MB-231细胞均有不同程度的抑制作用,复合组优于单一成分组,抑制生长效果与时间和剂量呈依赖关系;将人乳腺癌MB-231细胞阻滞于G0/G1期,抑制肿瘤细胞的有丝分裂;促进肿瘤细胞凋亡,与空白组相比差异显著。结论:复合螺旋藻多糖通过阻断细胞周期达到抑制体外培养的人乳腺癌MB-231细胞的生长的目的,具有较好的抗肿瘤活性。 Objective: To explore the inhibitory effects and possible mechanisms of compound polysaccharides from Spirulina Platensis (PSP)on human mammary cancer 231cells in vitro. Methods: Cells were treated with different concentrations of polysaccharides from Spirulina Platensis compounded with Ginkgo Biloba Extract ( GBE ) at different ratios. After 48h and 72h, the morphological changes of cells were observed under microscope, cytotoxic effects were evaluated by MTT method. After 48h cells were stained with PI and )AnnexinV -FITC/PI, the cell cycle and apoptosis ratio were measured by flow cytometry. Results : Compound polysaccharides from Spirulina Platensis of different ratio were cytotoxic to MB-231 cells in a time and dose dependent manner. And the inhibitory rates of them were higher than that of the single groups. The proportion of MB-231cells in GO/G1 phase was significantly increased. The mitosis of tumor cells was inhibited. The apoptosis rate of compound groups were remarkably increased compared with that in blank control group ( P〈0.05 ). Conclusion : Compound PSP can inhibit proliferation and increase apoptosis of MB-231 cells in vitro and has an ideal anti-tumor activity.
