摘要 背景与目的:Runx2在骨母细胞分化和骨形成中发挥重要作用,其在骨肉瘤中的作用尚不清楚,Ezrin是细胞骨架连接膜蛋白,与多种恶性肿瘤如乳腺癌、结直肠癌等转移相关。本研究旨在探讨骨肉瘤组织中Runx2与Ezrin基因的表达和相关性及其与临床生物学行为之间的关系。方法:应用免疫组化方法检测82例骨肉瘤活检样本石蜡组织中Runx2与Ezrin基因的蛋白表达,并分析其与骨肉瘤患者临床参数及生存之间的关系。结果:Runx2和Ezrin表达阳性率分别为65.9%和59.8%;Runx2和Ezrin的阳性表达与年龄、性别、部位、组织类型、ALP水平无关(P>0.05),而与肺转移相关,有肺转移者Runx2和Ezrin蛋白阳性表达率分别为100.0%和96.3%,显著高于无肺转移者(分别为49.1%和41.8%)(P<0.01);Runx2与Ezrin的表达呈显著正相关(P<0.01);Runx2阳性表达者和Runx2阴性表达者3年生存率分别为13.7%和64.3%,Ezrin阳性表达者和Ezrin阴性表达者3年生存率分别为12.5%和67.7%,Runx2和Ezrin阳性表达者预后较阴性表达者差(P<0.01)。结论:Runx2和Ezrin的表达与骨肉瘤肺转移相关;Runx2和Ezrin阳性表达水平可以作为判断骨肉瘤预后的指标。 Background and purpose: Runx2 plays an important role in the differentiation ofosteoblast and bone formation. Its role in tumorigenesis of osteosarcoma remains unclear. Ezrin is a membrane cytoskeletal linker protein and it is known to be associated with metastasis of many epithelial carcinomas such as breast and colorectal cancer, but its role in osteosarcoma also remains unclear. The purpose of this study was to investigate the expression ofRunx2 and Ezrin gene in osteosarcoma and its relationship with clinical parameters and to explore its potential clinical significance. Methods: The expressions of Runx2 and Ezrin were detected in 82 osteosarcoma biopsied paraffin- embedded samples by immunohistochemical method respectively. The relationships between Runx2, Ezrin and clinical parameters and survival of osteosarcoma were also analyzed. Results: The positive rate of Runx2 and Ezrin protein expression in osteosarcoma tissues were 65.9% and 59.8% respectively and the expressions of Runx2 and Ezrin had a significantly positive relationship to each other (P〈0.01). Its expressions were not related to clinical variables such as age, gender, site, histological type and ALP level (P〉0.05), but significantly related to lung metastasis (P〈0.01). Kaplan-Meier analysis revealed that osteosarcoma patients with Runx2 and Ezrin positive expressions had poorer prognosis than those with negative expressions of these two genes (P〈0.01). Conclusion: There is a relationship between Runx2 and Ezrin expression and lung metastasis of osteosarcoma. Both Runx2 and Ezrin expressions might be potential prognostic markers in osteosarcoma.
