大骨节病患者与健康人软骨细胞转化生长因子-β mRNA的表达差异 被引量:2

摘要 目的测定成人大骨节病(kaschin-beck disease,KBD)患者和正常健康成人关节软骨中转化生长因子-beta(transform growth factor-beta,TGF-β)在mRNA水平表达的差异,为进一步探讨大骨节病的发病机制奠定基础。方法提取成人大骨节病患者和健康人关节软骨细胞总RNA,逆转录得到cDNA;设计针对TGF-β基因的特异引物,通过RT-PCR技术检测该目的基因在mRNA水平表达差异。结果成人大骨节病患者的关节软骨细胞TGF-βmRNA表达低于健康成人。经统计学分析(t=11.1732,P<0.001),差异具有统计学意义(P<0.05)。结论成人大骨节病患者软骨细胞中TGF-β基因的表达低于健康成人,为进一步探讨大骨节病的发病机制和防治奠定了基础。 Objective To determinate the differences of transformation growth factor beta(TGF-β) mRNA expression in articular cartilage between Kaschin-Beck disease(KBD) patients and healthy people.Methods Total RNA was extract from articular cartilage from KBD patients and healthy adults.TGF-βgene expression was determined by RT-PCR.Results Expressions of TGF-β mRNA in articular cartilage cells of KBD patients was lower than that of healthy adult.And there was a statistically significant difference(P〈0.05).Conclusion Expression of TGF-β gene in cartilage cells of KBD adult patients was lower than that of healthy adults.
