呋喃唑酮代谢物AOZ在斑点叉尾体内组织分布与消除规律研究 被引量:9

摘要 研究了在池塘网箱养殖模式下,平均水温为30.6℃,以100 mg/kg鱼体重的剂量投饲斑点叉尾(Ietalu-rus punetaus)苗种(95±20.9)g含呋喃唑酮的药饵,连续5 d,每天2次。采用高效液相色谱串联质谱(HPLC-ESI/MS/MS)法分析了呋喃唑酮主要代谢产物3-氨基-2-噁唑烷酮(3-amina-2-oxazolidinone,AOZ)在斑点叉尾体内的组织分布与消除规律。结果表明:血液、肌肉、皮肤、肝脏和肾脏中AOZ于停药后在4 h时的浓度分别为(2 649.0±463.1)ng/mL,(1 169.4±194.5)、(2 308.5±100.5)、(7 816.2±568.5)和15 938.1μg/kg,血液、肌肉、皮肤、肝脏和肾脏中AOZ在60 d时平均浓度分别为6.66 ng/mL,4.50、33.7、17.1和21.3μg/kg,分别是AOZ判定限1μg/kg的6.66、4.5、33.7、17.1和21.3倍。90 d时AOZ在血液和各组织中均检测不到。AOZ在血液和各组织中的浓度水平肾脏>肝脏>皮肤>血液>肌肉,消除半衰期T1/2皮肤(11.1 d)>肌肉(7.8 d)>血液(7.2 d)>肝脏(6.9 d)>肾脏(6.7 d)。斑点叉尾皮肤中AOZ的浓度高于肌肉组织,而且残留时间长,在此养殖条件下,AOZ在斑点叉尾各组织中消除至少需要2 754℃.d。 Tissue distribution and elimination rules of furazolidone metabolite 3-amina-2-oxazolidinone(AOZ) was studied in channel catfish cultured in the cages of pond at the average temperature was 30.6 ℃ during the whole experiment.Channel catfish were continuously fed feed containing furazolidone twice everyday for 5 days at the dose of 100 mg/kg fish body weight.The concentrations of AOZ in blood and tissues(muscle,skin,liver,and kidney) were detected by liquid chromatography tandem mass spectrometric(HPLC-ESI/MS/MS).The concentrations of AOZ were(2 649.0±463.1)ng/mL,(1 169.4±194.5),(2 308.5±100.5),(7 816.2±568.5) and 15 938.1 μg/kg in blood,muscle,skin,liver,and kidney respectively after drug withdrawal for 4 hours.Average concentrations of AOZ in blood,muscle,skin,liver,and kidney were 6.66 ng/mL,4.50,33.7,17.1,21.3 μg/kg respectively on the 60th day and were 6.66,4.50,33.7,17.1,21.3 times of 1 μg/kg which was the determination limit of AOZ.AOZ was not detected on the 90th day in blood and all kinds of tissues.The concentration levels of AOZ from high to low in tissues were kidney,liver,skin,blood,and muscle.The elimination half-life T1/2(d) were 11.1 d(skin),7.8 d(muscle),7.2 d(blood),6.9 d(liver),and 6.7 d(kidney).The concentration of AOZ in skin was higher than in muscle and residue time was longer.In this cultured conditions,the elimination period of AOZ’ in channel catfish tissues was 2754 ℃·d.
