丹参多酚酸盐预处理对大鼠心肌缺血再灌注损伤的防护作用 被引量:28

摘要 目的:观察丹参多酚酸盐预处理对大鼠心肌缺血再灌注损伤的防护作用。方法:将健康成年雄性SD大鼠40只随机分为假手术组(S组)、缺血再灌注组(I/R组)、低剂量组(LD组)和高剂量组(HD组)。采用结扎大鼠左冠状动脉前降支(LAD)30min、恢复灌注120min制备大鼠心肌缺血再灌注模型。分别于药物输注前(T1),LAD阻断后30min(T2),LAD开放后30min(T3)、3h(T4)、6h(T5)、24h(T6)共6个时点测定血清一氧化氮合酶(NOS)、超氧化物岐化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GPX)、肌酸磷酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)及肌钙蛋白(cTnI)水平。结果:与I/R组比较,LD组和HD组血清CK-MB、LDH、cTnI、MDA及NOS水平显著降低,SOD与GPX含量显著上升(均P<0.05)。结论:丹参多酚酸盐预处理对大鼠心肌缺血再灌注损伤具有较好的防护作用。 Objective: To investigate the effect of salvianolate preconditioning on myocardial ischemia reperfusion injury in rats. Method:Healthy adult male SD rats were randomly divided into sham-operated group(S group),ischemia-reperfusion group(I/R group),low-dose group(LD group) and high-dose group(HD group).Myocardial ischemia and reperfusion was induced by ligating left anterior descending coronary artery(LAD) for 30 min and restoring perfusion for 120 min.We measured serum nitric oxide synthase(NOS),super oxide dimautase(SOD),malondiadehyde(MDA),glutathione peroxidase(GPX),reatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH) and troponin(cTnI) levels before infusion(T1),30 min after LAD ligation(T2),and 30 min(T3),3 h(T4),6 h(T5) and 24 h(T6) after LAD perfusion respectively. Result:Compared to I/R group,the levels of CK-MB,LDH,cTnI,MDA and NOS decreased significantly in LD group and HD group,while SOD and GPX elevated(all P〈0.05). Conclusion:Salvianolate preconditioning provided a protective effects on myocardial ischemia reperfusion injury.
