摘要
目的建立岗梅中冬青苷XXIX(ilexoside XXIX)的HPLC-ELSD含量测定方法。方法以PhenomenexBDS C18柱(250 mm×4.60 mm,5μm)为固定相,乙腈-0.2%甲酸为流动相,流速为1.0 mL.min-1,柱温为28℃,检测器为SEDEX 75型蒸发光散射检测器(漂移管温度为40℃,载气压力2.5 bar)。结果 IlexosideXXIX质量在0.984~29.52μg范围内的自然对数值与峰面积的自然对数值成良好线性关系,r=0.9998;平均回收率为100.8%,RSD为1.52%。测定不同批次市售岗梅药材的含量,显示Ilexoside XXIX在根和茎中的含量基本集中在0.1%~0.4%。结论该方法灵敏度高,重复性好,为岗梅药材的质量控制提供参考。
Objective To establish a HPLC-ELSD method for the determination of ilexoside XXIX in Ilex asprella. Methods Chromatographic conditions included Phenomenex BDS C18 column(250 mm×4.60 mm, 5 um), the mobile phase consisting of a mixture of acetonitrile-0.2 % formic acid. The flow rate was 1.0mL/min, the column temperature was 28 ℃, and the detector was SEDEX 75(Eva: 40 ℃, Gas flow: 2.Smbar) Results The linear range of ilexoside XXIX was 0.984-29.52 ug(r=0.9998). The average recovery was 100.8 %, RSD was 1.52 %, and the content in different hatches of Ilex asprella on the market was 0.1%-0.4 %. Conclusion The method is sensitive and with good repeatability, and it can provide reference for quality control of Ilex asprella.
