转化生长因子β1及丁酸钠对大肠癌细胞增殖的影响 被引量:3

摘要 目的:研究转化生长因子β1(TGF-β1)及丁酸钠对大肠癌HCT-116细胞体外增殖的影响,并探讨其可能的机制。方法:采用MTT比色法观察两药对大肠癌细胞增殖的抑制,ELISA检测其对尿激酶系统的影响,流式细胞仪(FCM)分析其对细胞周期的影响。结果:MTT显示TGF-β1与丁酸钠均能抑制HCT-116细胞的增殖,呈浓度和时间依赖性(P<0.05);两药联用可使其作用不同程度地增强。FCM显示两药均能使G1期细胞比例升高,与对照组比较差异均有显著性(P<0.05);两药联用可使其作用不同程度的增强。TGF-β1能够上调UPA活性和PAI-1水平(P<0.05),丁酸钠则抑制UPA活性(P<0.05),对PAI-1水平无明显影响。丁酸钠可部分抑制TGF-β1引起的UPA活性的增加(P<0.05),而对PAI-1无明显影响。结论:TGF-β1与丁酸钠均可以抑制大肠癌HCT-116细胞的增殖及阻止细胞周期的进展,两者联用具有协同作用;同时TGF-β1与丁酸钠可通过调节尿激酶系统影响大肠癌细胞的增殖与浸润。
