摘要
以植物锁阳(cynomorium songaricum rupr.)为材料,对其中水溶性多糖进行分离纯化、表征。锁阳粉末经过热水浸提、乙醇沉淀方法得到锁阳粗多糖。经过木瓜蛋白酶,Sevag法脱蛋白,H2O2法脱色,DEAE-纤维素离子交换柱层析,冷冻干燥,分离得到锁阳多糖CSP-D1组分,并用红外光谱(IR),热重-差热法(TG-DTA)进行了表征。咔唑法测定其糖醛酸含量为27.9%,凝胶渗透色谱法(GPC)测得分子量为5.5×104。经完全酸水解,衍生化,结合气相色谱-质谱法(GC-MS)测定了其单糖组成为鼠李糖、阿拉伯糖、甘露糖、葡萄糖、半乳糖;百分含量分别为:3.70%、15.15%、13.58%、44.26%、23.30%。研究表明CSP-D1是锁阳中新发现的一种多糖。
The crude polysaccharide was obtained from Cyrtomorium songaricum Rupr. by water extraction followed by ethanol precipitation. The CSP-D1 was obtained after removal of protein by Sevag and papain enzymolysis method, deeolour by H202, separated on DEAE-Cellulose and freezing vacuum drying. And the CSP-D1 was characterized by the methods of FT-IR spectra and thermo gravimetry-differential thermal analyses (TG-DTA). Uheriody, the contents of GAL-A was 27.9 % ,measured by carbazole method. And determination of the molecular weights of CSP-D1 by gel permeation in chromatography (GPC) was 5.5x104. Gas chromatography-mass spectrometry (GC-MS) indicate that the CSP-D1 was composed of Rhamnose (3.70 %), Arabinose(15.15 %), Mannose(13.58 %), Glucose(44.26 %) and Galactose(23.30 %).
