一株红树林真菌胞外多糖免疫增强作用研究 被引量:4

A Study on Immunomodulatory Effect of Exopolysaccharide from One Fungi Strain Isolated from Mangrove

摘要 对分离自海南红树林真菌菌株PH0016的胞外多糖(exopolysaccharide,EPS)免疫增强活性进行研究。从发酵上清中获取胞外多糖成份,体外实验以外周血单个核细胞刺激增殖实验观察EPS免疫增强活性;将菌株以50 mg/kg剂量连续灌胃小鼠28 d后,对小鼠进行体内免疫功能测定。同时根据形态观察和ITS序列分析对菌株进行鉴定。结果表明,相比于正常对照组,EPS在体外可有效促进外周血单个核细胞增殖(P<0.05)。EPS可有效提高胸脾指数、血清溶血素半数溶血值,增强迟发型超敏反应及巨噬细胞吞噬功能(P<0.01)。菌株经鉴定为淡紫色拟青霉。从海南红树林分离出一株淡紫色拟青霉,其胞外多糖有较好的免疫调节活性,值得进一步深入研究。 The immunoregulatory effect of fungi strain PH0016 isolated from mangrove in Hainan province was explored. The EPS of strain PH0016 was obtained from fermental supernatant. The immunological enhancement of EPS in vitro was evaluated by observing peripheral blood mononuclear cell (PBMC) proliferation. Every mouse were administrated with 50 mg/kg through stomach for 28 days. The indices of thymus and spleen, HCs0 value, delayed type hypersensitivity reaction (DTH), and phagocytic function of macrophage were used to investigate the effects of EPS on immune function in vivo.The strain was identified by means of morphological observations and analysis of the ITS sequence. Compared with the control group, EPS increased PBMC proliferation with dose of 5 mg/mL, 100 ~L (P〈0.05). EPS with dose of 50 mg/kg increased the indices of thymus and spleen and the HC50 value (P〈0.01), enhanced delayed type hypersensitivity reaction and phagocytic function of macrophage (P〈0.01). The strain was identified as Paecilomyces lilacinus. EPS from fungi strain PH0016 isolated from mangrove in Hainan province had remarkable a immunoregulatory effect, this EPS could be utilized for the further research.
