摘要
采用SDS法、简易提取法、高盐沉淀法和改良的CTAB法从油松(Pinus tabulaeformis)针叶提取基因组DNA,通过琼脂糖凝胶电泳、紫外分光光度法和PCR检测提取的DNA质量。结果表明,改良的CTAB法提取的油松针叶基因组DNA浓度和纯度高,适合用于ISSR-PCR分析,优于其他3种提取方法。
SDS method,simple extraction method,high salt extraction method and modified CTAB method were applied to extract DNA from the needle of Chinese pine(Pinus tabulaeformis).And the quality of extracted DNA was determined by agarose gel electrophoresis,UV spectrophotometry and PCR.The results showed that the concentration and purity of DNA obtained from modified CTAB method was better than that from the other 3 methods,and could be used in ISSR-PCR analysis of P.tabulaeformis.
