摘要
目的提取茵陈中的多糖,并进行含量测定。方法采用水提醇沉法提取茵陈多糖,以葡萄糖标准液绘制标准曲线,用苯酚-硫酸法、紫外分光光度法于484 nm波长处测定茵陈多糖的含量。结果多糖在484 nm有最大吸收峰,求得其回归方程为y=8.612 5x+0.065 2,相关系数r=0.999 8;平均回收率为98.4%,RSD=1.4%(n=5)。结论茵陈多糖含量为0.47%,该方法操作简便、重现性好、准确、可靠,可作为检测茵陈多糖含量的优选方法,为茵陈的质量控制提供了依据。
Objective To extract Artemisia capillaries polysaccharide, and to determine the content of poly-saccharide from Artemisia capillaries. Methods Water extracting and alcohol precipitation is applied to extract pol-ysaccharides from Artemisia .capillaries, with glucose as standard substance,using phenol-sulfuric acid method, for content determination of polysaccharides from Artemisia capillaries. Results Polysaccharide at 484 nm has maxi-mum absorption peak, its regression equation is y=8.612 5x + 0.065 2, the correlation coefficient is r=0. 999 8, and the average recovery rate is 98. 4%, RSD= 1.4 %(n = 5). Conclusion Artemisia capillaries polysaccharide content is 0.47%. The results show that the proposed method was convenient, economic, accurate and reproducible, and can be used to determine the polysaccharide in Artemisia capillaries, which provides a reference for the mass control of Artemisia capillaries.
