红细胞生成素对人白蛋白诱导肾小管上皮细胞转分化的作用 被引量:2

摘要 目的探讨红细胞生成素(EPO)对人白蛋白诱导的正常人近端肾小管上皮细胞(HK-2细胞)转分化的作用。方法将体外培养的HK-2细胞随机分为空白对照组(未加任何刺激物)、EPO诱导组(EPO终浓度为20 U/mL)、白蛋白诱导组(白蛋白终浓度5 mg/mL)、EPO干预组(EPO终浓度分别为5、10、20 U/mL)及转化生长因子β1(TGF-β1)拮抗剂组(SB431542终浓度为10μmol/L,加入SB431542 30 min后加白蛋白,白蛋白终浓度为5 mg/mL),各组均刺激24 h。应用RT-PCR法检测细胞TGF-β1 mRNA的水平;细胞免疫荧光法检测细胞E-钙黏蛋白(E-cadherin)、α-平滑肌肌动蛋白(α-SMA)的蛋白表达;ELISA法检测上清液纤维连结蛋白(FN)的蛋白表达。结果 RT-PCR结果表明白蛋白诱导组TGF-β1 mRNA较空白对照组显著升高(P<0.05),给予EPO干预后TGF-β1 mRNA显著下调(P<0.05);细胞免疫荧光及ELISA结果显示白蛋白诱导组E-cadherin的蛋白表达较空白对照组显著下降,α-SMA、FN的蛋白表达显著上升(P<0.05),而在EPO干预组及TGF-β1拮抗剂组E-cadherin的蛋白表达较白蛋白诱导组显著上调,α-SMA、FN的蛋白表达显著下调(P<0.05),且与EPO的浓度有关。结论 EPO可抑制白蛋白诱导的HK-2细胞的转分化,从而延缓肾间质纤维化,其机制可能与阻断TGF-β1的作用有关。
