不同光照形式对豚鼠视网膜色素上皮细胞TGF-β_2表达的影响

Effect of different forms of light on TGF-β2 expression of guinea pig retinal pigment epithelial cells in vitro

摘要 目的:探讨不同光照形式对豚鼠视网膜色素上皮(retinal pigment epithelium,RPE)细胞表达转化生长因子-β2(transforming growth factorβ2,TGF-β2)的影响。方法:幼年健康豚鼠(2周龄)10只,体外培养RPE细胞,传代、鉴定后,将细胞分为聚焦光组、离焦光组、平行光组和空白对照组,前3组分别接受聚焦光、离焦光(均为将平行光经透镜转化)和平行光照射,空白对照组不接受照射。于照射后24h采用免疫细胞化学法、实时荧光定量PCR法(real-time fluorescence quantitative PCR,RTFQ-PCR)检测细胞中TGF-β2及TGF-β2mRNA的表达,分析不同光照形式与效应的关系。结果:免疫细胞化学法显示各组TGF-β2表达均为阳性,病理VISTA图像分析软件测量单位面积中平均光密度(optical density,OD)值进行半定量分析,聚焦光组明显高于其它各组,组间比较有统计学差异(F=11.08,P<0.05);RTFQ-PCR法显示,聚焦光组TGF-β2mRNA表达水平较其它各组明显增加,组间比较有统计学差异(F=133.01,P<0.05)。结论:不同形式光线可影响豚鼠RPE细胞TGF-β2表达,以聚焦光最明显。 AIM: To investigate the effects of different forms of light on the expression of TGF-β2 of guinea pig retinal pigment epithelial(RPE)cells. METHODS: Ten two weeks old healthy guinea pigs, RPE cells were cultured conventionally and divided into four groups: focused light group, defocused light group, parallel light group and the control group. The first three groups were exposed to the focused light, defocused light(passed though lens from parallel light)and the parallel light, the control group was untreated. The level of the expression of TGF-β2 protein and TGF-β2mRNA was measured by immunocytochemistry and real-time fluorescence quantitative chain reaction 24 hours after light exposure and the pattern-effect relations were explored. The data were analyzed by means of analysis of variance(ANOVA). RESULTS: The level of both TGF-β2 protein and TGF-β2 mRNA were significantly increased in the focused light exposed group compared with the other groups, followed with the defocused group(P〈0.05).CONCLUSION: It was suggested that different forms of light can affect the expression of TGF-β2 in guinea pig RPE cells, and was most obvious in the focused light exposed group.
