摘要 目的通过对红细胞特异性δ-氨基γ-酮戊酸合成酶(5-aminolevulinic acid synthtase E,ALASE)转基因小鼠尿液代谢组学的研究,探究转基因小鼠尿液代谢组学上的变化及其意义。方法应用超高液相-四极杆-飞行时间/质谱检测分析系统,采用主成分分析和偏最小二乘-判别分析方法,筛选出差异性的物质。结果将3月龄的雌性转基因小鼠及其同窝阴性鼠的尿液进行聚类分析,发现了23种可能成为该小鼠模型特异性代谢产物。结论 ALASE转基因小鼠代谢状态通过代谢组学方法可以得到很好的体现,因而代谢组学技术为转基因小鼠的研究提供了良好的研究手段。 Objective To analyse the metabonome characteristics and significance of the urine of erythrocytes overexpression of ALAS mouse in order to explore metalSolomics of urine changes and'its meaning. Methods The metabonomics technique, UPLC-Q/TOF MS analytical means, principal components analysis (PCA)and (PLS-DA) were made in metabolic finger print data partial least squares discriminant analysis of different groups and different metabolites of porphyria mouse. Results After the UPLC-Q/TOF MS analysis, 23 potential discriminant metabolites of transgenic mouse and wild mouse were found. Conclusion The whole metabo- lism status of transgenic mouse model is able to be presented finely with the method of metabo- lomics. The metabolomics study could offer a satisfactory research approach to transgenic mouse.
