大豆苷元对异丙肾上腺素所致心肌肥厚大鼠血管活性物质的作用 被引量:4

Effect of daidzein on the changing of vasoactive substance in myocardial hypertrophy rats

摘要 目的:观察大豆苷元对异丙肾上腺素所致大鼠心肌肥厚的保护作用及其对血管活性物质的影响。方法:采用背部皮下注射异丙肾上腺素建立大鼠心肌肥厚模型。连续给药14 d,计算心脏重量参数,检测血浆血管紧张素Ⅱ、内皮素、心钠素、一氧化氮及6-酮-前列腺素F1α及心肌羟脯氨酸含量。结果:模型组心脏重量参数、心肌羟脯氨酸、血浆血管紧张素Ⅱ及内皮素含量升高,心钠素及一氧化氮含量降低;与模型组相比,0.1,0.3μmol/kg大豆苷元治疗组心脏重量参数、心肌羟脯氨酸、血浆血管紧张素Ⅱ及内皮素含量下降,血浆心钠素、一氧化氮及6-酮-前列腺素F1α含量升高。结论:大豆苷元对异丙肾上腺素所致大鼠心肌肥厚具有保护作用,其作用机制与升高扩血管物质心钠素、一氧化氮及前列腺素I2及降低收缩血管的活性物质血管紧张素Ⅱ和内皮素的水平有关。 Object: To observe the protection of daidzein (DD) on rat~ myocardial hypertrophy caused by isopropylarterenol (Iso) and explore the relationship of vasoactive substance. Method: The rat's myocardial hypertrophy model is formed through hypodermic injection which extended 10 days at back skin with Iso, 1 mg/kg. Once the model is formed, it will be administrated with various doses of DD (0.1,0.3 μmol/kg ), DMSO or NS through hypodermic injection from the second day, and the process will extend 14 days. After the last administration, the rat will be in absolute diet status for 12 h and its blood and heart are taken under anesthesia. The indexes of heart weight was calculated, the concentration of hydroxyproline in myocardium tissue was detected by alkali method, the level of angiotensin II (Ang I] ) , endothelin(ET) , atrial natriuretic peptide (ANP) and 6-keto-prostaglandin F1 a, (6-keto-PGFla) in plasma were investigated with radioimmunity kits, and the content of NO in serum was estimated by nitrate reductase assay. Result: There is obvious difference compare Iso group with control group. Indexes of heart weight and content of hydroxyproline of Iso group are much bigger than control one, level of ANP and NO are decreased while Ang I] and ET are decreased. However, indexes of heart weight is decreased in DD groups, furthermore, ANP, NO and 6-keto-PGF1 a level are enhanced while Ang lI and ET concentration are reduced in DD groups. Conclusion: DD has a protection effect on rat~ myocardial hy- pertrophy caused by Iso, and its mechanism probably result of modulating the concentration of vasoaetive substance such as Ang Ⅱ, ANP, ET, NO and 6-keto-PGFla.
