逍遥散对雌性兔膝关节软骨细胞超微结构的影响 被引量:1

Study on the effect of XIAOYAO POWDERS on ultrastructure of knee-joint chondrocytes in female rabbits

摘要 目的:探讨逍遥散对雌性兔膝关节软骨细胞超微结构的影响。方法:采用改良Huith法对30只雌性新西兰兔右膝关节进行膝骨关节炎造模,造模1周后将30只实验兔随机分为逍遥丸组、玻璃酸钠组及对照组,每组10只。逍遥丸组每天每只给予8丸逍遥丸(相当于原药材3 g),配成30 mL液体灌胃,连用5周;玻璃酸钠组予以玻璃酸钠注射液0.15 mL.kg-1右膝关节内注射,每周1次,连用5周;对照组每天予以30 mL生理盐水灌胃,连用5周。造模后12周末处死所有实验兔,取右膝内侧胫骨平台软骨组织切片后透射电镜观察。结果:逍遥丸组细胞核正常,核膜完整,核质均匀,线粒体嵴部分模糊,出现空泡化现象,内质网、高尔基体正常,纤维组织稍紊乱,有凋亡小体存在;玻璃酸钠组细胞核正常,核膜完整,核质均匀,线粒体空泡化较轻,内质网、高尔基体正常,纤维组织稍紊乱,有凋亡小体存在;对照组细胞核可见固缩现象,线粒体异常,表现为线粒体嵴模糊、消失,空泡化现象较明显,内质网、高尔基体基本正常,纤维组织紊乱现象较明显,多个标本见凋亡小体。结论:逍遥散对雌性兔膝关节软骨细胞具有保护作用。 Objective:To explore the effect of XIAOYAO POWDERS on the uhrastructure of knee-joint chondrocytes in female rabbits. Methods : Models of knee osteoarthritis (KOA) were built in right knee joints of 30 female New Zealand rabbits by the improved Huith meth- od. One week after modeling,30 experimental rabbits were randomly divided into XIAOYAO POWDERS group, sodium hyaluronate group and control group, l0 cases in each group. Every rabbit in the XIAOYAO POWDERS group was intragaustric administrated with 30 mL liquid which made from 8 XIAOYAO POWDERS( which dosage was equivalent to that of 3 g original medicine)every day for consecutive 5 weeks. Rabbits in the sodium hyaluronate group were injected with 0.15 mL · kg-1 sodium hyaluronate injection in their right knee joints once a week for consecutive 5 weeks,while the others in the control group were intragastrie administrated with 30 mL normal saline every day for consecutive 5 weeks. At the end of12 weeks afler modeling,all the experimental rabbits were executed,and sections from the tibial plateau cartilage tissue inside of their right knees were fetched out to be observed under transmission electron microscope. Results:Such situations were found in XIAOYAO POWDERS group as normal cell nucleus, complete nuclear membrane, uniform nueleoplasm, partially obscuring in mitochondrial crista,vacuolation,normal endoplasmie reticulum and Golgi' s apparatus, slightly disorder in fibrous tissue and existence of apoptotic bodies. Such phenomenons were found in sodium hyaluronate group as normal cell nucleus, complete nuclear membrane, uniform nucleoplasm,lesser vacuolatiun of initochondria, normal endoplasmic reticulum and Golgi' s apparatus, slightly disorder in fibrous tissue and existence of apnptotic bodies. The conditions appeared in a number of specimens of control group as pyenosis of cell nucleus, abnormal mito- chondria shown as blurred or disappeared mitochondrial crista, much more obviously vacuolation of mitochondria, essentially normal endo- plasmic reticulum and Golgi' s apparatus, much more obviously disorder in fibrous tissue and apoptotic bodies. Conelusion:XIAOYAO POWDERS has proteetive effect on knee-joint chondrocytes in female rabbits.
