摘要
目的研究经侧脑室移植骨髓间充质干细胞(mesenchymal stem cells,MSCs)对帕金森(Parkinson’s disease,PD)大鼠行为能力及中脑黑质TH表达的影响。方法雄性SD大鼠36只,随机分为假手术组(Sham组),PD模型组(PD组),MSCs移植组(MSCs组)。采用6-OHDA制备PD大鼠模型,经侧脑室移植骨髓MSCs,术后2周、4周、8周,腹腔注射阿朴吗啡诱导PD大鼠旋转行为,采用Western blot方法测定中脑黑质TH蛋白的表达。结果移植术后2周、4周、8周,MSCs组与PD组比较,旋转次数明显减少(P<0.05),移植细胞干预PD模型8周后中脑黑质TH表达,MSCs组较PD组明显增高(P<0.05)。结论经侧脑室移植骨髓MSCs通过提高中脑黑质TH的活性,改善PD大鼠的行为能力。
Objective To observe influence on expression of tyrosine hydroxylase (TH) in rats with Parkinson's disease PD treated by bone mesenchymal stem cells (MSCs). Methods The rat models of Parkinson's disease were set up by 6-OHDA and then divided into four groups randomly, which were Sham group, PD group, MSCs group. The rotating behavior of rat models induced by Apomorphine intraperitoneally with transplanting bone MSCs through cerebral lateral ventricle after 2, 4 and 8 weeks. TH protein in Mesencephalic nigra were measured by Western blot. Results The rotation numbers of PD group in the 2th, 4th and 8th week after transplanting cells were significantly increased compared with Sham group (P〈0.05), while that of MSCs sronp were significantly decreased compared with that of that of PD group. The levels of TH protein in nigra after PD model intervented by Cell transplantation after eight weeks were decreased significantly PD group compared with Sham group(P〈0.05), while that of MSCs were increased significantly compared with that of PD group. Conclusion The behavior of rats with Pakinson's disease was improved significantly by increasing the level of TH in nigra with transplanting BDNF modified bone MSCs through cerebral lateral ventricle.
