摘要
目的探讨前列腺素DP受体基因(PTGDR)启动子-549位点单核苷酸多态性(SNP)与哮喘的关系,观察基因型的分布频率。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)和琼脂凝胶电泳的方法检测哮喘组与健康对照组PTGDR启动子-549基因型及等位基因,计算该位点基因型和等位基因在哮喘组与健康对照组分布频率,然后运用χ2检验比较两组的基因型频率及等位基因频率有无显著差异。结果哮喘组与健康对照组PTGDR启动子-549位点有3种基因型,即CC型,CT型和TT型,哮喘组和健康对照组的3种基因型之间基因型出现频率比较有显著性差异(χ2=6.82,P=0.009),哮喘组与健康对照组T等位基因频率和C等位基因频率经比较两者有显著差异(χ2=6.820,P=0.033)。结论前列腺素DP受体基因(PTGDR)启动子-549位点单核苷酸多态性(SNP)与哮喘发病有一定的相关性。
Objective To explored the relationship of DP receptor gene(PTGDR)-549 promoter single nucleotide polymorphism(SNP) with asthma, and to observe distribution of genotype frequency. Methods Detected the protanoid DP receptor gene (PTGDR)-549 promoter of asthma group and healthy control group By polymerase chain reaction- restriction fragment length polymorphism(PCR-RFLP)and agarose gel electrophoresis, calculated the genotype frequencies and allele genotype frequencies distribution in the asthma grou and the healthy control group, then compared which by χ2 test.Results There were three gene types of PTGDR-549 promoter in asthma group and healthy control group:CC,CT and TT, and there were singnificant difference of the three gene types and frequencies in asthmatic group and healthy control group tested by χ2 test.(χ2=6.82,P=0.009),There were significant differences of T and C allele genotypes between the asthma group and healthy control group, (χ2=6.820,P=0.033).Conclusion The PTGDR-549 promoter single nucleotide polymorphism may correlated with asthma in the Han population.
