青藤碱对环瓜氨酸肽刺激类风湿关节炎T细胞分泌INF-γ及IL-4的影响 被引量:8

摘要 目的观察青藤碱对环瓜氨酸肽刺激的类风湿关节炎(RA)患者外周血T细胞分泌干扰素(INF)-γ及白细胞介素(IL)-4的作用。方法体外分离培养24例RA患者外周血淋巴细胞,分为空白对照组、环瓜氨酸肽组、甲氨蝶呤组及青藤碱组。体外培养72h后,用酶联免疫吸附法(ELISA)检测细胞培养上清中IFN–γ及IL-4水平。结果与空白对照组相比,环瓜氨酸肽组IFN–γ水平升高,差异有统计学意义;经青藤碱作用后,IFN-γ有所下降,差异具有统计学意义。与空白对照组相比,环瓜氨酸肽组分泌的IL-4降低,差异有统计学意义;经青藤碱作用后,IL-4明显升高,差异具有统计学意义。结论青藤碱调节环瓜氨酸肽刺激的RA患者外周血T细胞分泌IFN-γ及IL-4等细胞因子,可能在RA的治疗中发挥重要作用。 Objective: To observe effects of sinomenine on the secreting IFN-γ and IL-4 of T cell from rheuma- toid arthritis activated by cyclic citrullinated peptide. Methods: Peripheral blood lymphocyte(PBL) of 24 RA pa- tients were cultured in vitro. All bodies were divided into the blank control group,the cyclic citrulline peptide group,the methotrexate group and the sinomenine group. After cultured respectively in vitro for 72 h, the concen- trations such as IFN-5, and IL-4 were measured by ELISA. Results: Compared with the blank control group, con- centration of IFN-γ from cyclic citrulline peptide group was increased obviously. After being used the sinome- nine, the IFN-γ level was decreased significantly. The IL-4 in the cyclic citrulline group was less than that in the blank control group. However,its level was increased remarkably. Conclusion: The IL-4 and the IFN-γ are im- portant in the rheumatoid arthritis treatment which are activated by the cyclic citrullinated peptide adjusting by the sinomenine.
