摘要
目的建立超高效液相-三重四极杆串联质谱法(UPLC-MS/MS)测定黑小麦中维生素B1(VB1)、维生素B2(VB2)和维生素B6(VB6)的含量。方法用超高效液相色谱仪,ACQUITY UPLC BEH C18(2.1 mm×50 mm,1.7μm)色谱柱,以0.1%甲酸溶液和乙腈为流动相,进行梯度洗脱,在电喷雾(ESI)正离子模式下,用MRM模式监测。结果 VB1、VB2和VB6在20~200 ng/mL范围内线性关系良好。黑小麦中VB1、VB2和VB6的含量分别为0.965~1.961,0.614~0.963和0.364~0.625μg/g。结论该方法具有前处理简单、检测限低、回收率高的特点,适用于植物中VB1、VB2和VB6的定性、定量分析。
Purpose A novel method for determining the concentration of Vitamin B, (VB1) , Vitamin B2 ( VB2 ) , and Vitamin B6 ( VB6 ) isolated from black wheat was established using UPLC MS/ MS. Meth- ods The separation of VB1 ,VB2 ,VB6 was performed by using the ACQUITY UPLC BEH ClS column(2.1 mm ×50 mm,1.7μm). 0.1% formic acid and acetonitrile were used as the mobile phase. An electros- pray positive ion multiple reaction monitoring(MRM) was used as a detector. Results The study showed that the linear range of the determination failed between 20 and 200 ng/mL. Using this method,the con- centration of VB1 in black wheat was determined and the range of their concentration was 0. 965-1. 961μg/g,VB2 was 0. 614-0. 963 μg/g and VB6 was 0. 364-0. 625 μg/g · Conclusion The method estab- lished in this study is convenient, accurate and rapid with a lower detection limit,and suitable for the rap- id determination of VB1 ,VB2 and VB6 in plant.
