通心络对糖尿病大鼠心肌纤维化的防治作用 被引量:11

摘要 目的探讨通心络对糖尿病大鼠心肌纤维化的防治作用及机制。方法用链脲佐菌素制备大鼠糖尿病模型并给予通心络灌胃,12周后测定各组大鼠心脏重量指数,胶原容积积分,免疫组化测定心肌中转化生长因子-β1(TGF-β1)、Smad3、Smad7的蛋白表达水平,RT-PCR法检测TGF-β1、Smad3、Smad7mRNA的表达水平。结果糖尿病组大鼠心脏重量指数、胶原容积积分明显增高,TGF-β1、Smad3蛋白的表达增加,Smad7蛋白的表达减少;TGF-β1、Smad3mRNA的表达增加,Smad7mRNA的表达减少,通心络组上述指标均得到改善。结论通心络对糖尿病大鼠心肌纤维化有防治作用,其作用机制可能通过改善糖尿病大鼠心肌中TGF-β1、Smad3、Smad7的表达,从而减轻和延缓糖尿病大鼠心肌的纤维化有关。 Purpose To observe the effect of Tongxinluo on myocardial fibrosis in diabetic rats and speculate the possible mechanisms. Methods Rats were induced as diabetes mellitus models by given Streptozotocin(STZ). The diabetic rats were perfused Tongxinluo ultrafine powder into the stomach. After 12 weeks,the heart weight index were calculated. Myocardial collagen volume fraction(CVF) were meas- ured by image analyzer. The mRNA and protein levels of TGF-[31 , Smad3 , Smad7 were detected with RT- PCR and immunohistochemistry respectively. Results The heart weight index and the CVF values in- creased significantly,the protein and mRNA levels of TGF-β1, Smad3 increased, the protein and mRNA levels of Smad7 decreased in the model group. The changes of the heart tissue had improved in the Tongx- inluo group. Conclusion Tongxinluo can prevent and treat the myocardial fibrosis in diabetic rats. It can lessen and delayed diabetes myocardial fibrosis. The possible mechanism is to improve the expression lev- els of TGF-β1 ,Smad3, Smad7 mRNA and protein.
