重组降血压肽缓释微球对自发性高血压大鼠的降压作用 被引量:1

摘要 目的考察重组降血压肽聚乳酸(rAHP-PLA)缓释微球对自发性高血压大鼠(SHR)的降压作用。方法采用口服和皮下注射两种给药方式,分别给予按体质量和血压均衡分组的自发性高血压大鼠rAHP-PLA缓释微球,空白组给予等量生理盐水。结果口服给予降血压肽缓释微球后10 h,自发性高血压大鼠的血压降至最低,且药效可以持续36 h;皮下注射rAHP-PLA缓释微球后30 h,血压降至最低,且可以持续约9 d。rAHP-PLA缓释微球对正常血压无影响。结论 rAHP-PLA缓释微球具有显著的降压效果,并有明显的缓释作用。 Purpose To study the effect of biodegradable microspheres loaded with recombinant anti- hypertensive peptide (rAHP) on blood pressure of spontaneously hypertensive rats. Methods SHR were randomized into ten groups ( n = 8, each group) and received one of the following approches by gavage and subcutaneous injection respectively : saline, rAHP ( 800 μg/kg ) , low-dose ( 200μg/kg ) , medium-dose (400 μg/kg) and high-dose(800 μg/kg) of the rAHP-PLA microspheres. Thirty-two WKY rats were ran- domized into four groups (n = 8, each group), and rAHP-PLA microspheres (800 μg/kg) and saline were administrated by garage and subcutaneous injection respectively. The systolic blood pressure was meas- ured befo,',~ ^-:'~ .'~tc, the administration. Results The results showed that compared with the "control group,medium-dose and high-dose of rAHP-PLA microspheres reduce blood pressure after being orally administrated and the effect can last for above 36 hours. For the subcutaneous injection groups, medium- dose and high-dose of rAHP-PLA microspheres reduce blood pressure and the effect can last for above 9 days. rAHP-PLA microspheres had no effect on reducing blood pressure in WKY rats. Conclusion The an- ti-hypertensive effects of the rAHP-PLGA microspheres persist for much longer time,with no effect on nor- motensive WKY rats. Conclusion The anti-hypertensive effects of the rAHP-PLA microspheres persist for much longer time,with no effect on normotensive WKY rats.
