摘要
目的探讨兔肝VX2移植瘤模型建立和肿瘤发展过程中血清及瘤组织S100A6蛋白表达的动态变化,为阐明其在肿瘤发展过程中的作用积累实验依据。方法建立兔肝VX2移植瘤模型,观察期自接种第7天至第29天。ELISA法检测血清中S100A6的水平,SP免疫组织化学法检测组织中S100A6的表达。结果大体和病理检查证实模型建立成功,成功率为100%(49/49)。ELISA检测发现,对照组的血清S100A6均值为(4.09±0.20)ng/mL;荷瘤组各观测点的S100A6水平均低于对照组,第17、19、21、23、25、27、29天依次较对照组减少37.6%、79.9%、89.9%、92.9%、98.3%、98.5%和99.0%,差异显著(P<0.05),且减少趋势也具显著性(P<0.05)。在各转移癌灶中,S100A6的表达也随荷瘤进程进行性降低趋势(P<0.01)。结论在兔肝VX2移植瘤模型的肿瘤发展过程中,兔血清和肿瘤组织中S100A6的表达均呈进行性降低。提示S100A6蛋白血清含量可能成为肝癌分期和是否转移的标志物;恢复S100A6的表达可能对兔肝VX2移植瘤的发展和转移产生重要作用。
Objective To explore the dynamic changes of S100A6 protein expression both in serum and transplanted tumor of S100A6 in rabbit VX2 carcinoma,and to explain the contribution of S100A6 in tumor events. Methods Establish rabbit hepatic VX2 transplanted tumor hepatic tumor model and to inspect S100A6 expression both in serum (ELISA) and tumor tissues (immunohistochemistry SP) through a range from 7th day to 29th day. Results Rabbit hepatic VX2 transplanted tumor hepatic tumor model has been established successfully ( 100% ). Experimental group concentration of S100A6 in serum are all lower than the level of control group, and decreasing amplitudes are 37.6% ,79.9% ,89.9% ,92.9% ,98.3% ,98.5% ,99.0% respectively from the 17th day (P 〈0.05) ,and the decreasing trend is obvious ( P 〈 0.05 ). A stepwise decrease positive stain is observed in tumor tissues ( P 〈 0.01 ). Conclusion S100A6 is involved in rabbit hepatic VX2 transplanted tumor hepatic tumor and shows a stepwise decreasing manner both in serum and tumor tissue which predicts. Serum concentration of S100A6 may indicate tumor grade and metastasis as a marker. Longitudinal experiments on recovering expression of S100A6 may affect metastasis.
