摘要
目的观察抑郁症对大鼠结肠组织中神经肽Y(neuropeptide-Y,NPY)和降钙素基因相关肽(calcitonin gene related peptid,CGRP)含量的影响。方法按照Open-Field行为学评分方法,选取20只Wistar大鼠,随机分成对照组和抑郁模型组,利用长期不可预见的温和应激刺激建立大鼠抑郁模型。采用放射免疫方法分别检测大鼠结肠组织中NPY和CGRP的含量。结果与正常组比较,模型组大鼠的垂直运动、水平运动和糖水消耗量均减少,提示建模成功;结肠组织中NPY的含量显著降低[模型组(4.93±1.72)ng/g、正常组(13.49±4.76)ng/g,P<0.01];CGRP的含量显著增加[模型组(19.82±6.05)ng/g、正常组(8.43±3.32)ng/g,P<0.01]。结论抑郁症可能通过影响大鼠结肠组织中NPY和CGRP的释放而导致大鼠结肠的功能异常。
Objective To investigate the effects of depression on the contents of brain-gut peptides neuropeptide-Y(NPY) and calcitonin gene related peptide(CGRP) in colon tissues of rats.Methods 20 wistar rats were by Open-Field behaviors and randomly divided into normal group and depression group.The chronic mild stimulations were used to produce classic depression model of rats.The concentrations of NPY and CGRP in colon of rats were detected by radioimmunoassay.Results The rearing,crossing and sucrose-consuming of depression group were decreased significantly.The data suggested the model was established successfully.The concentration of NPY in colon tissues of depression model rats was decreased significantly compared with control group [model group(4.93±1.72)ng/g,control group(13.49±4.76)ng/g,P〈0.01],while the CGRP level in colon tissues of depression model rats was increased significantly compared with normal control [model group(19.82±6.05)ng/g,control group(8.43±3.32)ng/g,P〈0.01].Conclusion The release of NPY and CGRP in colon tissues may be related to the abnormality of colon presented in depression model of rats.
