摘要
目的通过国产DNATyperTM15和进口IdentifilerTM试剂盒对陈旧骨骼、牙齿和指甲样本的检验结果比对分析,了解国产试剂盒对降解检材的检验效果,探讨在案件检验中应用的可行性。方法提取陈旧骨骼、牙齿和指甲样本的DNA后,用DNATyperTM15及IdentifilerTM试剂盒同步扩增,并用检出率、杂合子基因座上等位基因均衡性(PHR)和等位基因分型结果一致性,对DNATyperTM15和IdentifilerTM试剂盒进行比较。结果 45例陈旧骨骼、牙齿和指甲样本,DNATyperTM15试剂盒成功检出399例,检出率88.7%,IdentifilerTM试剂盒成功检出404例,检出率89.8%;DNATyperTM15试剂盒和IdentifilerTM试剂盒共同检出398例,在这398例样本中,PHR≤0.5分别为128个和117个;同一样本中,相同基因座上两种试剂盒检出的等位基因分型结果完全一致。经统计学分析,两种试剂盒的检出率和杂合子基因座上等位基因均衡性的检测结果均无统计学差异(P>0.05)。结论国产DNATyperTM15试剂盒可有效应用于实际案件中陈旧骨骼、牙齿及指甲样本的检验。
Objective Through the comparative analysis of domestic DNATyper^TM15 and imported Identifiler^TM kit for old bones,teeth and nails samples test results,understanding of the test results of degradation of samples with domestic kit,and investigating the feasibility of the application in test cases.Methods After extracting old bones,teeth and nail samples of DNA,amplifying them synchronously with the domestic DNATyperTM15 and imported Identifiler^TM kit,and with the detection rate,PHR of allele on heterozygote locus,and consistency of allele typing results,comparing the domestic DNATyper^TM15 and imported Identifiler^TM kit.Results In 450 cases of old bones,teeth and nail samples,DNATyper^TM15 kit successfully detected 399 cases,the detection rate is 88.7%,Identifiler^TM kit successfully detected 404 cases,and the detection rate is 89.8%.DNATyper^TM15 kit and Identifiler^TM kit jointly detected 398 cases,in the 398 cases of common samples,PHR ≤ 0.5 were respectively 125 and 117.In the same sample,typing results of allele detected by both kits on the same locus were in agreement.After statistical analysis,both detection rate of the two kits and the test results of PHR of allele on heterozygote locus were not statistically different(P〈0.05).Conclusion Domestic DNATyper^TM15 kit can be applied effectively in actual cases inspecting old bones,teeth and nails samples.
