摘要
在水稻雄性不育系花粉细胞败育程度鉴定方法上,传统的单纯显微图像分析方法或纯光谱分析方法存在定性、定位、定量和定时四位一体分析能力不足的问题。本文在普通荧光显微镜的基础上,研制出一种基于LCTF(Liquid Crystal Tunable Filter,液晶可调谐滤光器)的显微光谱成像分析系统,并将其引入到水稻花粉发育过程中的细胞学定量研究,初步建立起一套针对水稻花粉透射光光谱图像分析和荧光光谱图像分析新方法。与传统的纯光谱或纯图像分析方法相比,本分析方法具有独特的定性、定量、定位和定时四位一体的分析能力,其对水稻雄性不育系花粉败育程度的鉴定将更为准确。本方法对于其它植物和动物细胞的定量分析同样具有借鉴和推广意义。
For pollen rice male sterility identity techniques,traditional pure microscopic image analysis or spectral analysis is not enough for qualitative,quantitative,locating and timing analysis.On the basis of normal fluorescence microscope,a LCTF(Liquid Crystal Tunable Filter) device based microscopic spectral imaging system was constructed and introduced to the quantitative study for rice pollens.A set of transmitted light and fluorescence spectral image analysis techniques were developed.Compared with traditional pure spectral or image analysis method,it has the capability for simultaneously qualitative,quantitative,locating and timing analysis,and thus more precise results could be obtained.This method is promising for other plants and animals quantitative analysis in cellular level.
