6-羟基多巴胺对星形胶质细胞hepcidin表达的影响 被引量:1

EFFECTS OF 6-HYDROXYDOPAMINE ON hepcidin mRNA EXPRESSION IN ASTROCYTES

摘要 目的观察6-羟基多巴胺(6-OHDA)处理对星形胶质细胞hepcidin mRNA表达水平的影响。方法应用实时定量PCR方法,检测原代培养的星形胶质细胞经6-OHDA处理6h及24h后hepcidin mRNA表达水平的变化,以无血清培养液培养的细胞为对照组。结果与对照组比较,6-OHDA处理6h及24h的星形胶质细胞中hepcidin mRNA表达水平均显著下降,差异有统计学意义(F=6.116,q=4.314、4.467,P<0.05);而6h和24h处理组间差异无统计学意义(q=0.017,P>0.05)。结论氧化应激状态下星形胶质细胞中的hepcidin可能参与细胞内的铁代谢异常。 Objective To investigate the expression of hepcidin mRNA in astrocytes treated by 6-hydroxydopamine(6-OHDA).Methods By employing real time PCR,the expression of hepcidin mRNA in primarily cultured astrocytes treated by 6-OHDA for six or 24 hours was detected,and the cells cultured with serum-free medium were served as controls.ResultsCompared with the controls,the expressions of hepcidin mRNA levels in 6 h and 24 h-6-OHDA-treated astrocytes mRNA were down-regulated,the differences being significant(F=6.116;q=4.314,4.467;P〈0.05);but the difference between that of 6 h and 24 h-treatment was not(q=0.017,P〈0.05).Conclusion Under the state of oxidative stress,hepcidin in astrocytes might involve in intracellular iron metabolism disorders.
