摘要
目的研究急性呼吸窘迫综合征(ARDS)小鼠液体复苏时给予外源性清蛋白对肺血管通透性和含水量的影响。方法 144只昆明种小鼠,随机分为正常对照组和致伤组,致伤组分为未治疗组、体积分数0.1清蛋白治疗组、体积分数0.2清蛋白治疗组。采用脂多糖(LPS)15mg/kg腹腔注射制备小鼠模型,应用改良伊文思蓝渗出法测定模型制备后4、8、16、24h各组肺组织伊文思蓝渗出量及肺湿质量/干质量(W/D)比值的变化。结果各致伤组小鼠各时间点肺组织伊文思蓝渗出量与正常对照组比较差异均有显著性(F=2 124.232~8 642.934,q=64.601~176.626,P<0.05)。各致伤组小鼠各时间点肺W/D比值与正常对照组比较,差异均有显著性(F=32.434~173.547,q=9.601~27.651,P<0.05)。与未治疗组和体积分数0.1清蛋白治疗组比较,体积分数0.2清蛋白治疗组肺W/D比值明显减小(q=2.865~9.897,P<0.01)。结论体积分数0.2清蛋白能降低ARDS小鼠早期肺含水量,不增加肺血管通透性,有助于ARDS早期的治疗。
Objective To investigate the effect of exogenous albumin on pulmonary capillary permeability(PCP) and lung water content in mice with acute respiratory distress syndrome(ARDS).Methods This study consisted of 144 Kunming mice,which were randomly divided into two groups: normal control and injured groups,and those in the injured group were divi-ded into untreated group,0.1-albumin-treated group,and 0.2-albumin-treated group.A model of ARDS in mouse was created by intraperineal injection of lipopolysaccharide(LPS,15 mg/kg).Evan's blue content in lung tissue and wet /dry(W/D) weight ratio were determined by modified Evan's blue method.Results Compared with the normal control group,the amount of Evan's blue effusion in the model mice increased significantly(F=2 124.232-8 642.934;q=64.601-176.626;P〈0.05).In the injured groups,the W/D ratio in all time points was significantly different as compared with the normal control(F=32.434-173.547;q=9.601-27.651;P〈0.05).Compared with untreated and 0.1-albumin-treated group,the W/D ratio in lung tissue of 0.2-albumin-treated group was dramatically lower(q=2.865-9.897,P〈0.01).Conclusion 0.2 albumin can reduce lung water content in early ARDS,and not increase PCP,which is conduce to the early therapy of ARDS.
