人端粒酶逆转录酶预测肺癌化疗敏感性的体外研究 被引量:4

摘要 目的:通过丙氧鸟苷(GCV)、5-氟尿嘧啶(5-FC)两种前体药物对肺癌细胞的杀伤,研究不同剂量化疗药物与hTERT表达水平间的关系。方法:采用不同浓度化疗前体药物5-FC、GCV对肺癌细胞刺激,提取细胞的总RNA,逆转录成cDNA后,用RealTime PCR测定各细胞hTERT mRNA的表达量。结果:5-FC和GCV不同药物浓度刺激A、B细胞后,hTERT mRNA表达量均下降,并随着5-FC浓度的增加,A、B细胞内hTERT mRNA的表达逐渐下降,呈负相关性;但GCV浓度在(100~1 600)μg/ml刺激后,A、B细胞内hTERT mRNA表达相同。结论:hTERT与化疗敏感性的关系,与抗肿瘤药物、细胞本身敏感性以及hTERT有关。 Objective:To study the relationship between hTETR expression and different dose of chemotherapeutics by investigating the cancer-killing effect of GCV and 5-Fu on lung cancer cells.Methods:Two lung cancer cell lines were treated with different concentration of 5-FC and GCV,the total RNA was then collectd and reverse transcripted to cDNA.The expression of hTERT mRNA was evaluated by RealTime PCR.Results:After treatment with different concentration of 5-FC and GCV,the lung cancer cell lines were decreased expression in hTERT mRNA.There was a negative correlation between 5-FC concentration and hTERT mRNA expression.However,after tumor cells treated with 100~1 600μg/ml of GCV,the expression of hTERT mRNA was increased.Conclusion:Antitumor drug,sensitivity of tumor cell was involved in the correlation between hTERT and chemosensitivity.
