摘要
以黑曲霉为出发菌株,通过紫外诱变,筛选得到一株产β-呋喃果糖苷酶活力较高的突变株YA03,其产酶活力是出发菌株的1.66倍,且遗传稳定性良好。采用单因素和响应面法对该突变株产β-呋喃果糖苷酶的发酵培养基进行优化,得到最佳产酶培养基:蔗糖11.86%、牛肉膏0.53%、MnCl2.4H2O 0.14%、MgSO.47H2O 0.05%、FeSO.47H2O 0.05%,此条件下β-呋喃果糖苷酶活力可达96.58 U/ml。
Aspergillus niger for the original strain,the mutation strain YA03 producing high β-fructofuranosidase activity was screened by UV mutagenesis,its enzyme activity is 1.66 times than the original strain with genetic stability.The mutagenesis strains producing β-fructofuranosidase of fermentation medium used univariate and response surface method to optimize the best medium:sucrose 11.86%,beef extract 0.53%,MnCl2·4H2O 0.14%,MgSO4·7H2O 0.05%,FeSO4·7H2O 0.05%.Under these conditions,β-fructofuranosidase activity reached 96.58 U/ml.
