摘要
目的检测microRNA-199a-3p(miR-199a-3p)在肾癌细胞株和组织中的表达情况并探究miR-199a-3p在肾癌细胞中的作用。方法利用实时定量RT-PCR检测miR-199a-3p在肾癌细胞和组织中的表达水平;利用miR-199a-3p模拟物转染肾癌细胞786-0上调miR-199a-3p后,通过CCK-8、克隆形成、Transwell以及细胞周期检测来探究其在肾癌细胞中的作用。结果 miR-199a-3p在肾癌细胞中明显低表达,在78%(14/18)的肾癌组织中亦明显低表达;上调miR-199a-3p可显著抑制肾癌细胞的增殖、存活和侵袭并能诱导细胞周期G1期阻滞。结论我们的研究显示在肾癌中miR-199a-3p明显低表达并参与肾癌的发生、发展,这表明miR-199a-3p具有作为肾癌诊断和治疗靶点的潜能。
Objective To study the expression of microRNA-199a-3p (miR-199a-3p) in renal cell carcinoma (RCC) cell lines and tissues, and assess the impact of miR-199a-3p on the tumorigenesis of RCC. Methods miR-199a-3p levels in RCC cells and tissues were assessed by real-time qRT-PCR. The RCC cell line 786-0 was transfected with miR-199a-3p mimics to restore expression of miR-199a-3p. The effects of miR-199a-3p were then assessed by CCK-8, cell cycle, Transwell, and colony formation assay. Results We found that miR-199a-3p was remarkably downregulated in RCC cells and tissues. Restoration of miR-199a-3p dramatically suppressed RCC cells growth, clonability, invasion and induced G1-phase cell cycle arrest in vitro. Conclusions Collectively, these results demonstrated that miR-199a-3p was dramatically downregulated in RCC and involved in the etiology of RCC, which suggested that miR-199a-3p may offer an attractive new target for diagnostic and therapeutic intervention in RCC.
