摘要
目的:探讨自噬相关基因Beclin1和微管相关蛋白LC3表达和腮腺肿瘤之间的关系。方法:用免疫组化方法检测20例正常腮腺组织、37例腮腺多形性腺瘤、41例癌在多形性腺瘤中Beclin1和LC3的表达,并结合临床病理因素进行分析。结果:Beclin1在正常腮腺组织、腮腺多形性腺瘤、癌在多形性腺瘤中的阳性率分别为100%、92%、63%;LC3在这3种组织的阳性表达率分别为100%、86%、56%。淋巴结转移及TNM分期与Beclin1的表达间有显著性关系(P<0.05),不同侵袭性、有无淋巴结转移与LC3的表达间有显著性关系(P<0.05)。结论:Beclin1和LC3在腮腺多形性腺瘤及癌在多形性腺瘤中表达下调,自噬功能改变可能在腮腺肿瘤形成中具有一定作用。
Objective: To investigate the expression of microtubule-associated protein light 1 chain 3 (LC3) and Beclinl in parotid tumors. Methods: The expression of Beelinl and LC3 was assessed by immunohistochemistry in 20 cases of normal parotid gland ( NPG), 37 cases of parotid pleomorphic adenoma(PPA) and 41 cases of carcinoma in pleomorphic adenoma(CPA). The correlations of Beclinl and LC3 expression to the clinicopathologic characteristics of CPA were analyzed. Results: The positive rates of Beclinl in NPC, PPA and CPA were 100% ,92% and 63% respectively, and those of LC3 were 100% , 86% and 56% respectively. In CPA, the expression of Beclinl was associated with lymph node metastasis and TNM stage( P 〈 0.05 ) , the invasive of the tumor and lymph node metastasis was associated with the LC3 expression( P 〈 0.05 ). Conclusion : The expressions of LC3 and Beclinl are down-regulated in PPA and CPA. The changes of autophagic capacity may be related to the development of.parotid gland tumors.
