摘要
采用大孔树脂法纯化从野生石蒜中提取的石蒜总碱,用高速逆流色谱法进行分离,用高效液相色谱法进行检测。结果表明,高速逆流色谱法分离石蒜碱和加兰他敏的最佳溶剂体系为:正己烷∶乙酸乙酯∶甲醇∶水=3∶7∶3.5∶6.5(体积比),分离后石蒜碱的纯度为95.36%,加兰他敏的纯度为91.67%。高速逆流色谱法可以同时将石蒜总碱中石蒜碱和加兰他敏分离,分离效果良好。
Macroporous resin was applied to purify alkaloids from Lycoris. The alkaloids was separated using high-speed counter-current chromatography (HSCCC) and was detected using high performance liquid chromatography. The results showed that the optimal solvent system for HSCCC to separate lycorine and galanthamine from Lycoris was hexane-ethyl acetate-methanol-water(3 : 7 : 3.5 : 6. 5, volume ratio). The purity of lycorine reached 95.36 % and the purity of galanthamine reached 91.67 % after separation. The high-speed counter-current chromatography was proved to be effective to separate lycorine and galanthamine simultaneously.
