摘要
目的:评价Nutlin-3对紫外线诱发的人黑素细胞DNA氧化损伤的保护作用。方法:aMSH预处理MClR功能正常的人黑素细胞,分组予以紫外线照射(UVB,105 mJ/cm^2)、Nutlin-3(10μM)和UVB+Nutlin-3不同干预方法,并采用彗星实验确定各组DNA托尾率、尾距和尾矩数值。结果:UVB组DNA托尾率、尾距和尾矩数值高于其他各组(P<0.05),UVR+Nutlin-3组明显低于UVB组(P<0.05)。结论:Nutlin-3能够减少UVB诱导黑素细胞DNA损伤。
Objective: To evaluate the protective effects of Nutlin - 3 on oxidative DNA damage of the melanocytes after ultraviolet exposure. Methods: Normal functioning MC1R melanocytes were pretreated with a - MSH, followed with ultraviolet irradiation (UVB, 105 mJ/cm2 ), Nutlin - 3 ( 10 t^M) and UVB plus Nutlin - 3, respective- ly. Tailing rates, tail movements and tail torque of each group were analyzed with Comet Assay. Results: Tailing rates, tail movements and tail torque were the highest in the UVB group ( P 〈 0.05), and were significantly lower in the UVB + Nutlin - 3 group than in the UVR group ( P 〈 0.05). Conclusion: Nutlin - 3 can reduce melanocytic DNA damage induced by UVB irradiation through a non- melanogenesis route.
