摘要 目的探讨不同提取工艺对提取蚕砂中叶绿素及制取叶绿素衍生物脱镁叶绿酸的影响,以提高蚕砂叶绿素及叶绿素衍生物的制备量。方法在单因素试验基础上,按L9(33)正交设计考察影响叶绿素提取率的主要因素,按C16(44)正交设计考察影响制备脱镁叶绿酸a出率的主要因素。结果蚕砂软化时间2 h(含水量26%)、丙酮-乙醇(1∶2,V/V)做溶剂为提取叶绿素的最佳条件,提取率提高到1.37%。以蚕砂叶绿素粗品合成叶绿素衍生物脱镁叶绿酸,浓盐酸脱镁搅拌反应72 h,制备率达67%。结论新工艺提高了蚕砂叶绿素及其衍生物脱镁叶绿酸的提取制备率。 Objective To explore the influence of different extraction technologies on the extraction of chlorophyll and chlorophyll derivatives from silkworm excrement in order to improve the amount of them. Methods The extraction technology of chlorophyll from silkworm excrement was studied with L9(33) orthogonal experiment after the single factor experiments. The effect of magnesium removal time with hydrochloric acid on yield of pheophorbide from chlorophyll were studied with C16(44) orthogonal experiment. Results The optimized extraction technology of chlorophyll from silkworm excrement was as follows: the softening time of silkworm excrement was 2 h (26% of water content), and the extraction solvent was acetone-ethanol (1 : 2, V/V). The production rate of chlorophyll was 1.37%. The preparation parameters of pheophorbide from chlorophyll were removing magnesium with hydrochloric acid for 72 hours. The yield of pheophorbide was 67%. Conclusion The rate of production of chlorophyll and pheophorbide from silkworm excrement increased by improving extraction technology.
