摘要 MEN1基因是多发性内分泌肿瘤1型综合征的关键致病基因之一,其编码蛋白为menin。研究显示menin在转录调节、基因组稳定、细胞分裂增殖及细胞周期调控中扮演着重要角色。在转录调控过程中,menin与JunD、C-jun、核因子κβ等大量转录因子及DNA双链直接或间接相互作用调控着众多靶基因表达,同时menin可以和组蛋白修饰酶相互作用在表观遗传学层面上调节基因的转录。Menin与大量涉及基因组稳定的蛋白相互作用,影响着基因组的稳定,在缺乏menin表达的细胞中基因组DNA极不稳定。Menin还与大量涉及细胞分裂增殖及细胞周期调控的蛋白相互作用,共同调控着细胞增殖及细胞周期。 M enin, the protein encoded by the gene MENI,is the key disease-causing protein for multiple endocrine neoplasia type 1 syndrome. As a conservative protein, menin had been observed to be involved in various cell functions, such as gene transcription, maintenance of genomic integrity, cell division control and cell differentiation. Menin can directly or indirectly regulate numerous gene transcription in different man- ners,such as interacting with transcription factors JunD and C-Jun, NF-KB to regulate genes expression, or binding double strand DNA. Menin also influences gene transcription on epigenetic level by interplaying with histone modifying enzymes. Menin also plays a key role in genome stability through interacting with DNA re- pair associated proteins. Genome DNA instability can be observed when there is no menin expression. Menin can also regulate cell proliferation and cell cycle control by interacting with lots of related proteins.
