黄连温胆汤对实验性大鼠慢性胃炎模型EGF及bFGF表达的影响

Experimenal Study of Decoction of Huang-lian Wen-dan on Expressions of EGF and bFGF in Chronic Gastritis

目的:以黄连温胆汤为代表的清热除湿降逆法,干预实验性慢性胃炎模型SD大鼠,观察胃窦黏膜的病理变化及对EGF、bFGF表达的影响。方法:把SD大鼠随机分为空白对照组,模型对照组,黄连温胆汤高、中、低剂量组,西沙比利组。分别给予相应干预对比观察胃黏膜改变,并进行胃窦部的病理切片,进行镜下观察,运用免疫组化方法检测上皮生长因子表达的影响。结果:胃窦黏膜炎细胞总面积和水肿总面积、炎症区域平均光密度、积分光密度,与模型组比较,西沙必利组、黄连温胆汤中、高剂量组程度均低,有统计学意义(P<0.05);与西沙必利组比较,黄连温胆汤高剂量组炎症明显降低,有统计学意义(P<0.05)。西沙比利组与黄连温胆汤高剂量组免疫组织化学比较,两者间无明显差异(P>0.05)。各组造模后的EGF、FGF表达较高,经药物干预后,EGF、bFGF的表达较疾病模型组升高,有统计学意义(P<0.05)。以黄连温胆汤高剂量组作用明显,组间比较无显著性差异(P>0.05)。结论:黄连温胆汤能够显著升高模型大鼠胃窦黏膜bFGF的表达,协同EGF促进局部血管的生成。

Objective ： To observe the effect of Decoction of Huang-lian Wen-dan , which was with the method of clearing away heat and eliminating dampness to calm the adverse-rising energy, on the gastric mucosal patholog-ical change and the expressions of EGF and bFGF in SD rat of chronic gastritis with the immunohistochemical method. Methods： The SD rats were divided into six groups, which were blank control group, model control group, high, midium and low dose of treatment group and control group of cisapride. After treated with related methods, pathological section of sinus ventriculi was observed under light microscope, and the expression of EGF and bFGF were detected with immunohistochemical method. Results： Compared with model control group the total area and edema area of gastric antrum mucous membranitis and average optical density and score optical density of inflammatory area were lower in control group of cisapride, medium and high dose of treatment group, which showed statistical difference （P 〈 0. 05 ）. Compared with control group of cisapride inflammation was lessened in high dose of treatment group, which also showed statistical difference （ P 〈 0. 05 ）. But there was not statistical difference on immunohistochemistry between control group of cisapride and high dose of treatment group （P 〉 0. 05 ）. After made into model the expressions of EGF and bEGF were higher in each group. And after interfered in by medicine they were increased more than in model control group, and showed statistical difference （P 〈 0. 05）. This difference was more significant in high dose of treatment group, but there was not significant differ-ence between the groups （ P 〉 0. 05 ）. Conclusion ： Decoction of Huang-lian Wen-dan can increase more markedly the expression of bFGF and promote local angiogenesis in coordination with EGF.
