肾安颗粒对ROD模型大鼠骨组织FGF-23表达及骨代谢的影响 被引量:4

摘要 目的探讨ROD模型大鼠骨组织成纤维细胞生长因子23与骨代谢的变化及中药复方肾安颗粒干预骨代谢作用机制。方法应用5/6肾切除加高磷饮水建立大鼠ROD模型,随机分为正常对照组、假手术组、模型组、骨化三醇组、肾安2.25g/ml剂量组、肾安4.5g/ml剂量组,给予相应药物灌胃8周,给药前、后检测大鼠血尿素氮、血肌酐、Ca、P、碱性磷酸酶,化学发光法测全段甲状旁腺素,双能X射线测骨密度,Westernblot、免疫荧光检测股骨成纤维细胞生长因子23表达。结果肾安2.25g/ml、肾安4.5g/ml组大鼠血尿素氮、血肌酐与骨化三醇组、模型组比较明显降低。肾安2.25g/ml、肾安4.5g/mlY/IJ量组血Ca、骨密度与模型组比较明显升高,血P、碱性磷酸酶、全段甲状旁腺素与模型组比较明显降低。各组大鼠骨组织成纤维细胞生长因子23蛋白表达,模型组显著高于正常组和假手术组,骨化三醇、肾安2.25g/ml、肾安4.5g/ml组明显低于模型组。肾安2.25g/ml、肾安4.5g/ml组免疫荧光成纤维细胞生长因子23表达较模型组明显减低。结论ROD模型大鼠骨代谢异常与成纤维细胞生长因子23异常表达密切相关,肾安颗粒对骨代谢影响机制可能与干预成纤维细胞生长因子23表达有关。 Objective To probe into the effects of Shen'an granule on the expression of FGF-23 and bone metabolism of ROD model rat osseous tissue. Methods The ROD model rats were randomly divided into 6 groups., normal group, sham operation group, model group, ealcitriol group, Shen'an Granule high dose group(4. 5 g/ml) and Shen' an Granule low dose group (2. 25 g/ml). 8 weeks' medication was needed, and BUN, SCr, serum calcium, serum phosphor, alkaline phosphatase, iPTH, bone density and FGF-23 were observed before and after each treatment in each group. Results Com- pared with the model group and calcitriol group, both Shen'an Granule high dose group and Shen'an Granule low dose group's blood urea nitrogen and serum creatinine were decreased significantly. Com- pared with model group,Shen'an Granule high dose group and Sherran Granule low dose group "s the se- rum calcium and bone density were significantly increased and serum phosphor, alkaline phosphatase, iPTH were decreased significantly. The expression of FGF-23 protein of the model group was signifi- cantly higher than that of the normal group and sham operation group. Both Shen'an Granule high dose group and Shen'an Granule low dose group were significantly lower than the model group. Conclusions Abnormal bone metabolism and abnormal expression of FGF-23 are closely related in the ROD model rats. It is likely that the effect of Sherran granule on the bone metabolism has relationship with the ex pression of FGF-23.
