摘要 目的研究核转录因子-κB(NF-κB)特异性抑制剂吡咯烷二硫代氨基甲盐酸(PDTC)对人黑素瘤A375细胞的生长抑制作用。方法不同浓度的PDTC作用于A375细胞不同时间后,MTT法测定其对A375细胞增殖的抑制率,免疫细胞化学方法检测PDTC作用下A375细胞增生细胞核抗原(PCNA)的表达情况。结果经PDTC处理的实验组,肿瘤细胞增殖活性明显受到抑制(P<0.05,P<0.01),呈时间和剂量依赖性;与对照组相比,PDTC作用后A375细胞PCNA表达降低(P<0.01),且呈明显的剂量依赖关系。结论 PDTC具有显著抑制人黑素瘤A375细胞生长及PCNA表达的作用,是一种有潜力的抑制黑素瘤增殖的药物。 Objective To explore the effect of pyrrolidine dithiocarbamate(PDTC),a potent inhibitor of NF-κB,on the proliferation of human melanoma cell line A375.Methods A375 cells were respectively treated with PDTC by different concentration and different time in vitro.The inhibition rates of cell growth were assayed by MTT method.Immunocytochemical method was used to analyze expression of proliferating cell nuclear antigen(PCNA).Results The tumor cells proliferation activity of experimental group with PDTC processing was inhibited obviously in a time-dependent and dose-dependent manner(P0.01).Compared with the control group,the expression of PCNA was down regulated in PDTC group in a dose-dependent manner(P0.01).Conclusions PDTC can inhibit the growth and PCNA expression of A375 cells,it may be a potential chemotherapeutics for the proliferation of melanoma.
