转抗真菌基因水稻几丁质酶和β-1,3-葡聚糖酶活性的时间变化 被引量:4

Temporal Changes of Both Chitinase and β-1,3-glucanase Activities in Transgenic Rice with Extra Anti-fungal Genes

摘要 为了研究外源几丁质酶基因和β-1,3-葡聚糖酶基因在水稻植株上表达的时间变化规律,利用酶促反应结合3,5-二硝基水杨酸(DNS)比色法测定了4个转基因水稻株系不同生育期叶片中几丁质酶活性和β-1,3-葡聚糖酶活性。结果表明,无论是亲本还是转基因株系,2种酶活性随生育期的后延而增大。生育前期,转基因株系的2种酶活性与亲本的酶活性水平差异小,随生育期的后延,与亲本酶活性的差异越来越大,说明外源基因前期表达量小甚至沉默或抑制内源基因的表达,后期表达量多。同时转基因株系的酶活性的波动性较亲本的大,且酶活性的高低与转入基因数量无相关性。因此,外源基因的表达具有时效性但不具有数量优势。 In order to study the temporal change of extra-genes express in rice, the activities of chitinase and β-1,3-glucanase in the leaves of the four transgenic rice lines with extra anti-fungal genes (chitinase gene and β-1,3-glucanase gene) in different growth period were measured using enzymatic reaction combined 3, 5-dinitrosalicyclic acid (DNS) colorimetry method. The results showed that the activities of chitinase and β-1, 3-glucanase in both transgenic rice lines and their parents increased with the proceeding of growth period. In the early growth period, there were slight difference of enzyme activities between transgenic rice lines and their parents, whereas the difference became greater with plant growth, which suggested that, in early growth period, there were low extra-genes express, even gene silencing or gene suppression. The curves of enzyme activities in transgenic lines varied greater than parents, and there were no correlation between enzyme activities and the number of extra gene. It was concluded that, for extra-gene express, there were time efficiency but no numerical superiority.
