摘要
目的初步探讨白颖苔草提取物抗人巨细胞病毒(human cytomegaloviras,HCMV)的作用机制。方法通过不同时间点干预HCMV—HEL感染细胞模型,确定提取物是在病毒入侵细胞前或后发挥抗病毒作用;应用RT—PCR检测即刻早期基因UL122、早期基因UI朋、晚期基因UL86mRNA的转录表达,探讨提取物发挥抗病毒作用的时段;通过观察HCMV—UL32荧光病毒在HEL增殖过程中的荧光信号变化,确定提取物是在HCMVUL32基因特录前或者转录后发挥作用。结果无论将提取物在病毒入侵细胞前、后进行干预HCMV—HEL感染模型,均可抑制细胞病变的发生;RT-PCB.结果提示干预模型的HCMVUL122、UL44基因的结果为阳性,而晚期基因UL86的转录表达为阴性;提取物可有效抑制荧光病毒的荧光信号的表达。结论白颖苔草提取物抗HCMV的作用机制主要是抑制中晚期的基因转录,而对病毒的入侵过程和即刘早期、早期基因的转录表达无明显影响。
Objective To investigate the mechanism of anfiviral activity of extract from Carex rigescens against human cytomegalovirus (HCMV). Methods Human embryo lung (HEL) fLbroblast cells were infected with human cytomegalovirus (HCMV) AD169 strain and HCMV - HEL cell model was established to determine whether the extract from Carex rigescens could play an antiviral activity before or after the in- fection by HCMV. The changes of fluorescent signals of HCM~ -UL32, a fluorescent virus, during the proliferation d ltEL were observed to determine whether the extracts can exert their effects before or after HCMV - UL32 gene transcription. Meanwhile, the mRNA transcription levels of immediate early gene (IE) UL122, early gene (E)UL44, and late gene (L) UL86 were detected by reverse transcription polymerase chain reaction ( RT - PCR) to determine the effect stage of the extract from Carex rigescens during the viral replication of HCMV. Results The cytopathogenic effect induced by the HCMV in the HCMV r HEL cell model was suppressed before and after the infection of the virus. In this study, ? the? fluorescence signal of HCMV - UL32 during the proliferation of HEL was suppressed by the extract from Carex rigescens. RT - PCR analyzation revealed that the transcription expressions of HCMV - ULI22 and HCMV - UL44 were positive while the expression of late gene HCbIV - UL86 was negative. Conclusions The mechanism of antiviral acti-dty of extract from Carex rigescens against HCMV based mainly on the suppression of transcription of viral late genes of HCMV ADI69 strain. The extract from Carex rigescens had no significant inlluence on the invasion of HCMV into HEL, the transcription of immediate early genes and late genes of human cytomegalovirus.
