摘要
目的:构建一种新型纳米抗体靶向MRI造影剂,负载超顺磁性四氧化三铁(superparamagnetic iron oxide,SPIO)聚合物胶束。方法:采用两亲聚合物包覆法制备负载SPIO的聚合物胶束,并检测该聚合物胶束的粒径、形貌、顺磁性氧化铁的含量、磁化曲线;用免疫方法检测聚合物胶束连接抗体的情况、普鲁士蓝染色法观察铁在前列腺癌细胞中铁标记率。结果:合成的负载SPIO的抗体靶向聚合物胶束的粒径为38.1±0.4;靶向负载SPION的聚合物胶柬中SPIO的含量为35.8%,电镜下负载SPION的靶向聚合物胶束成球形,粒径小且每个胶束里只有一二个SPIO粒子;功能化后的负载SPIO的性能并没有减弱;通过免疫荧光法证实前列腺干细胞抗原(PSCA)抗体连接在聚合物胶束,能促进了前列腺癌细胞对负载SPIO的抗体靶向聚合物胶束的吸收。结论:成功合成PSCA抗体靶向功能的SPIO聚合物胶束,在体外均显示出较好的靶向效果,可通过进一步的研究探索作为一种活体应用的MR/探针来检测前列腺癌。
Objective : to construct a new nano antibody targeted MRI contrast agents, load super paramagnetic ferroferfic oxide ( superpammagnetic iron oxide, SPIO ) polymer miceUes. Methods : the two amphipbilic polymer coating method for preparing supported SPIO polymeric mleelles, and detection d the polymeric mlcelles, particle size, morphology, superparamagnetie iron oxide content, magnetization curve; detection by immune method of polymeric micelles connected antibody, Prussian blue staining observation Fe AR in prostate cancer cells labeled rate. Results: synthetic load SPIO antibody targeting of polymeric micellar particle diameter was 38. 1± 0.4 ; target load SPION polymeric micelles in the content of SPIO is 35.8% , under the electron load SPION targeting polymer mlcelle into a spherical shape, small size and each miceUe in only one or two of the SPIO particle ; function of load the performance of SPIO does not diminish ; by immunefluorescence confirmed prostate stem cell antigen ( PSCA ) antibody is attached in polymeric micelles, can promote prostate cancer cells to load SPIO antibody targeting polymer miceUe absorption. Conclusion: the successful synthesis of PSCA antibodies targeting function SPIO polymeric micelles, in vitro showed better targeting effect, through further study and exploration as an in rive applica- tion of MRI probes for the detection of prostate cancer.
