氨基酸分析仪法测定角膜接触镜护理液中甘氨酸的含量

Determination of Glycin in Contact Lens Care Solution by Amino Acid Analyzer

目的建立用氨基酸分析仪测定角膜接触镜护理液(简称"护理液")中甘氨酸(Gly)含量的方法。方法采用日立氨基酸分析仪,甘氨酸经离子交换色谱茚三酮柱后衍生,以外标法进行测定。流动相:pH=4.0的柠檬酸钠缓冲液-0.2mol.L-1氢氧化钠溶液梯度洗脱,流速:0.4mL.min-1,检测波长:570nm。结果甘氨酸出峰时间为3.2min,整个分析时间仅为10min。甘氨酸在0.0107～0.214g.L-1范围内线性关系良好,相关系数为0.9999。加样回收率为100.1%～100.7%,RSD为0.16%～0.30%。结论该方法操作简便,无需样品前处理,准确度高,且护理液中其他成分对测定影响小。

Objective To determine Glycin （Gly） in a contact lens care solution by amino acidanalyzer. Methods Gly was analyzed by ninhydrin postcolumn derivative ion exchange chromatography method, using standard Gly solution as the external standard. The mobile phase was pH= 4.0 sodium citrate buffer solution and 0.2 mol ·L^-1 sodium hydroxide solution with gradient elution at a flow rate of 0.4 mL·min^-1 and a detection wavelength of 570 nm. Results The retention time of Gly was 3.2 rain, the whole analysis time was 10 rain. The linear correlation of Gly was 0. 0107-0. 214 g· L^-1 with correlation coefficient of 0. 9999. The average recoveries at three spiked levels were in the range of 100.1%-100.7% with relative standard deviations of 0.16%-0. 30%. Conclusion The method is accurate, simple and rapid for determination of Gly in contact lens care solution.
