粒细胞集落刺激因子对急性辐射损伤小鼠胸腺输出功能的影响 被引量:1

G-CSF promotes murine thymic emigration after a sublethal dose of irradiation

摘要 目的粒细胞集落刺激因子(granulocyte colony-stimulating factor,G-CSF)是急性辐射损伤治疗的首选药物,但其在促进粒系造血恢复的同时能否促进辐射损伤后中枢免疫功能恢复尚不清楚。文中以亚致死量辐射损伤小鼠为研究对象,观察G-CSF对急性辐射损伤后胸腺输出功能的影响。方法雌性BALB/c小鼠50只给于6.0 Gy60钴一次性全身照射后随机均分为2组。G-CSF组小鼠给予重组人G-CSF 100μg/(kg.d)皮下注射,连续14 d;对照组小鼠给予等体积磷酸盐缓冲液(PBS)皮下注射,连续14d。照后30d、60d分别利用流式细胞仪及荧光定量PCR技术检测外周血中初始T淋巴细胞比例以及胸腺细胞中T细胞受体重排删除环(T cell receptor rearrangement excision circles,TRECs)拷贝数,判断胸腺输出功能。结果照后30 d,G-CSF组小鼠外周血中初始T淋巴细胞比例G-CSF组(3.5±0.9)%与对照组(1.3±0.8)%比较差异有显著性统计学意义,P≤0.05;胸腺信号结合T细胞受体删除环(signal joint T cell receptor rearrangement excision circles,sjTRECs)拷贝数G-CSF组(6450±783)均明显高于对照组(3630±595),P<0.05。但照后60 d 2组上述检测指标差异无统计学意义。结论 G-CSF可在急性辐射损伤后早期增加胸腺输出功能,促进中枢免疫重建。 ZHAO Hong-xia1,GUO Mei2,SUN Xue-dong2,LIU Tie-qiang2,ZUO Hong-li2,LIU Guang-xian2,AI Hui-sheng2(1.Department of Hematology,General Hospital of the Second Artillery,Beijing 100088,China;2.Department of Hematology,The Affiliated Hospital of the Academy of Military Medical Sciences,Beijing 100071,China)
