摘要 目的:探讨血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)、环氧化酶-2(COX-2)和p-KDR在血管瘤中的表达及作用。方法:采用免疫组织化学SP法、实时定量PCR及Western免疫印迹法检测血管瘤标本、血管畸形标本及正常皮肤组织中VEGF、COX-2和p-KDR的表达,应用SPSS 13.0软件包对数据进行统计学分析。结果:免疫组织化学检测发现,VEGF、COX-2和p-KDR在血管瘤标本中高表达,其阳性率分别达100%、93.33%和86.67%,且与血管畸形和正常皮肤组织中的表达有显著差异(P<0.05)。实时定量PCR及Western免疫印迹方法检测发现,VEGF、COX-2和p-KDR mRNA及蛋白在血管瘤中的表达显著高于正常皮肤组织。结论:VEGF、COX-2和p-KDR在血管瘤血管内皮细胞上高表达,其相互作用可促进血管瘤增殖。其表达情况为临床治疗血管瘤提供了新的思路。 PURPOSE:To investigate the role of VEGF,COX-2 and p-KDR in dermal hemangiomas.METHODS: By using SP,real-time PCR and Western blot method,the expression of VEGF,COX-2 and p-KDR was observed in hemangiomas,vascular malformations and normal skins.Statistical analysis was performed using SPSS 13.0 software package.RESULTS:The expression of VEGF,COX-2 and p-KDR was high in hemangiomas,the positive rate was 100%,93.33% and 86.67%,respectively.There was significant difference between hemangiomas and vascular malformations and normal skins(P0.05).Real-time PCR and Western blotting showed the expression of mRNA of VEGF,COX-2 and p-KDR in hemangiomas was significantly higher than that in the normal skins.CONCLUSION: The expressions of VEGF,COX-2 and p-KDR in hemangiomas was significantly higher and their interaction could stimulate the development of hemangioma.This could provide a new route to manage hemangiomas.
