摘要
从土壤样品中分离获得一株产复合酶的真菌菌株,将其命名为HKS11,进行了菌株形态特征观察,并在此基础上进行了分子生物学鉴定,经PCR扩增后测定菌株的18S rDNA基因测序,由18S rDNA基因序列比较分析,得知菌株HKS11与黑曲霉菌(Aspergillusniger)和泡盛曲霉(Aspergillus awamori)亲缘关系最为接近,18S rDNA基因相似性达到99%以上,可初步判断为曲霉属(Aspergillus)。结合形态特征观察综合比较分析后,将其鉴定为曲霉属(Aspergillus)的黑曲霉菌(Aspergillus niger)。其18S rDNA GenBank中登录序列号为GenBankJX112703。并对其发酵产酶进行了研究。
Abslxaet: A fungi strain HKS 11 with high yield of compound enzyme was isolated from soil in the laboratory. Based on morphological characteristic, the sequencing 18S rDNA was done after PCR amplification. By comparison of 18S rDNA sequences, the isolated strain HKSI 1 was most similar to Aspergillus niger and Aspergillus awamori. The max ident of 18S rDNA was more 99%. By comprehensive and comparative analysis, HKSI I was identified as Aspergillus niger. Its accession number is JX 112703. It was studied on producing enzymes in 100L fermentator by HKS11.
