Correlation Study and Expression of Matrix Metalloproteinase-3 and Tissue Inhibitor of Metalloproteinase-3 in Human Colon Cancer

摘要 探讨基质金属蛋白酶-3(MMP-3)及抑制物组织金属蛋白酶抑制剂-3(TIMP-3)在人结肠癌组织中的表达及其意义。本文采用免疫组化的方法对86例结肠癌,40例结肠腺瘤和40例正常结肠黏膜组织中的MMP-3和TIMP-3的表达进行检测。结果MMP-3在结肠癌组织中的阳性表达率高于结肠腺瘤和正常结肠组织,差异有统计学意义(P<0.05);TIMP-3在正常结肠组织中的阳性表达率高于结肠癌组织和结肠腺瘤,差异有统计学意义(P<0.05)。结肠癌组织中MMP-3和TIMP-3蛋白的表达与结肠癌患者性别和年龄无关(P>0.05);与癌细胞分化程度、有无淋巴结转移、有无远处器官转移及临床分期密切相关(P<0.05)。MMP-3与TIMP-3蛋白在结肠癌中的表达呈负相关(P<0.05)。TIMP-3和MMP-3表达程度及TIMP-3/MMP-3可为结肠癌的浸润转移提供诊断依据,并为临床治疗结肠癌的浸润转移提供新的思路和方法。 To study the expressions of matrix metalloproteinase {MMP--3) and tissue inhibitor metalloproteinase (TIMP --3) in human colon cancer. Immunohistochemisty was used to detect the different expression of MMP--3 and TIMP--3 in 86 cases with colon tumor 40 cases of polypous adenoma tissues and 40 normal colons from incisal edges. MMP--3 in colon cancer tissue of positive expression rate is higher than colonic adenoma and the normal colonic organization, the difference was statistically significant { P〈0.05 ), TIMP--3 in normal colon tissue of positive expression rate is higher than colon cancer organizations and colonic adenoma, the difference was statistically significant (P〈0.05). Expression of MMP--3 and TIMP--3 protein in colon cancer had not significantly correlation respectively with patient's gender or age, (P〉0.05), but had significantly correlation with dukes stage, lymph node metastasis, and differentiation of cancer cell (P〈0. 05). There was a negative correlation between MMP--3 and TIMP--3expression (P〈0.05). The expressions of MMP--3 and TIMP--3 as well as the ratio of MMP--3 /TIMP--3 can be thought of colon cancer diagnosis based on transfer invasion embellish provide, and for clinical treatment of colon cancer assault embellish transfer to provide new train of thought and method.
